Towards a Green Macrocyclization Protocol: Development of Novel Strategies and Catalysis

Abstract: Our group has been developing novel techniques for the synthesis of macrocycles. Novel conformational control elements have been demonstrated to access rigid, chiral, well-defined macrocycles and cyclophanes. More recently, we have made progress towards developing general approaches for controlling dilution effects in macrocyclization via a “phase separation” strategy. The “phase separation” strategy has been applied to important contemporary chemical transformations commonly used to construct macrocycles, such as olefin metathesis, Glaser-Hay couplings and “Click” cycloadditions. Efforts to develop new catalytic strategies to access macrocycles via direct macrolactonization or Sonogashira cross-coupling will also be presented.
